3' RACE: skewed ratio of specific to general PCR primers improves yield and specificity.

چکیده

target sequence is correct, the target sequence is amplified, and if the primer pair is in the wrong orientation relative to the amplified cDNA inserts, the target sequence is not amplified. This simple, linear pre-amplification of cDNAs in the library has the potential for wide use. This approach requires not only far fewer starting materials than conventional PCR cloning but also circumvents lengthy conventional screening. Another important advantage is that it only requires one genespecific primer to clone cDNA sequence. Even when only one specific primer sequence is known, full-length cDNA can be amplified using the double-stranded sequence of one specific primer. A sense primer and an antisense primer from the same region can be used in separate PCRs in conjunction with a vector primer. Here we have shown that the simple amplification of a cDNA library by PCR can improve the resulting target bands. This is a very easy way of amplifying a cDNA library with a reduced risk of losing materials.
